IN VITRO SYNERGISTIC ACTIVITY OF ITRACONAZOLE AND SIX DIFFERENT PUMPS BLOCKERS AGAINST 16 CLINICAL ISOLATES OF ASPERGILLUS FUMIGATUS RESISTANT AND SUSCEPTIBLE TO ITRACONAZOLE

Abstract: 

Aim: to evaluate the activity for the combination of itraconazole with blockers drugs against A.fumigatus strains resistant and susceptible to itraconazole.MIC's were determined by broth microdilution method according to NCCLS guidelines (M38P) The checkerboard technique was used to study the interaction between itraconazole (ITZ) and the blockers. The final concentration ranged from 16 to 0.03 mg/l for ITZ; 640 to 10 mg/l for verapamil (VER); 8 to 0.13 mg/l for amiodarone (AMD); 16 to 0.25 mg/l for lidocaine (LID); 40 to 0.6 mg/l for lansoprazole (LAN); 8 to 0.13 mg/l for amiloride (AML) and 80 to 1.25 mg/l for fluphenazine (FLU). Final inoculum: 0.4 - 5 x 104 CFU/ml. The microtitration plates were incubated at 35oC and read after 48 h. The MIC's were defined as the lowest concentration of the drugs (alone or in combinations) that showed 50% of inhibition of the growth compared with the drug-free control. Drug interaction effects were defined on the Fractional inhibitory concentration index (FIC) Synergistic: 0.5 - 1 - 2.Synergistic action was found between itraconazole and some blockers as LAN, AMD, AML and FLU
