Transcription Analysis of Murine Kidney and Brain in Response to Systemic Aspergillosis.

Abstract: 

Background: Invasive Aspergillus fumigatus infection represents a challenge for immunocompromised patients. We performed expression profiling to analyze the transcriptional host-response in the kidney and brain of mice infected with A. fumigatus. Methods: CD-1 mice were infected iv with 8 x 106 conidia. Brain and kidneys were taken at 14 h postinfection from 3 uninfected and 3 infected mice and on d 5 and 9 from infected mice; these were frozen in liquid N2 prior to RNA extraction. cDNA was hybridized on murine genome-wide arrays MOE 430, 2.0 (Affymetrix). Expression analysis was performed using Array Assist 4.0 and Ingenuity. Results: In total, in kidneys, 181 genes (1 d), 4524 genes (5 d) and 3758 genes (9 d) and in brains, 28 genes (1 d), 383 genes (5 d) and 1783 genes (9 d) were differentially regulated.
