FR901379, A novel Antifungal Antibiotic.

Abstract: 

FR901379 and its related compounds have been isolated from the culture broth of Coleophoma empedri F-11899. They are water-soluble echinocandin-like lipopeptides with a sulfonare. They have the inhibitory activity on / beta-1,3-gluten synthase and exhibited in vivo activity against C.albicans, A.fumigatus and P.carinii in mice. The IC50 of FR901379 on C.albicans beta-1,3-glucan synthase was 0.7 ug/ml, whereas that of echinocandin B was 2.6 ug/ml. The IC50s of FR901379 against C.albicans FP633 and A.fumigatus FD050 in a micro-broth dilution test were 0.025 and 1.9ug/ml, respectively. It showed an exellent efficacy in a mouse C.albicans systemic infection model at the ED50 of 2.7 mg/kg by parenterel treatment. The prophylactic administration of FR901379 on P.carinii infested-nude mice completely eradicated the cysts in the lung.
