Abstract:
Novel antifungal agents are required to expand the antifungal armamentarium. Because of long development timelines we must be screening and searching now for drugs to be used in the clinic in 10 years time. Conventional microbiological screening still has a place in drug discovery but compound library selection is essential to provide novelty and diversity in the hits and compounds developed. The use of genomics to identify novel fungal targets led to various target based screening strategies which have largely been unsuccessful. Fragment based screening using carefully selected novel fungal targets may be a more successful approach as fragment hits are more amenable to development into drugs. In silico modelling and docking studies can be used alone or with existing techniques to assist and refine drug design and enable scaffold hopping. Using a combination of old and new techniques provides excellent opportunities for developing novel antifungals for the future.
