Phylogenetic analyses on the population diversity of Aspergillus fumigatus sensu lato based on CaM and cnB gene sequences

Abstract:

Ninety-eight isolates of Aspergillus fumigatus sensu lato mainly from China, as well as from
Australia, France, Germany, India, Indonesia, UK, and US were analysed to infer the population diversity
and haplotypes based on calmodulin gene (CaM), calcineurin regulatory subunit B gene (cnB)
sequences and mating types. The results from these strains showed that A. fumigatus sensu lato
contained one cryptic species, i.e., A. neoellipticus, while A. fumigatus sensu stricto consisted of two
populations, i.e., A. fumigatus population and A. phialiseptus population, in which, A. neoellipticus
consisted 5 haplotypes, A. fumigatus population included 9 haplotyopes, and A. phialiseptus population
had 10 haplotypes. Moreover, irrespective of species or populations, MAT1-1/MAT1-2 ratio was nearly
1:1 when clone-corrected, but when not clone-corrected, the ratio of MAT1-1/MAT1-2 was so biased as
near 1:2 in A. fumigatus sensu stricto. Furthermore, no correlation was found between populations or
haplotypes and antifungal drug tolerance.
