Abstract:
Dermatophytes are keratinophilic fungi able to infect keratinized tissues of human or animal origin. Among them,
Trichophyton mentagrophytes is known to be a species complex composed of several species or variants. Since the T.
mentagrophytes complex includes both anthropophilic and zoophilic pathogens, accurate molecular identificationis a critical
issue for comprehensive understanding of the clinical and epidemiological implications of the genetic heterogeneity of this
complex. Molecular strain typing tender an alternative way to identify each of fungal strains which will be very useful for
epidemiological purpose. Here, 47 archived T. mentagrophytes isolates were taken for the study. The isolates were confirmed
by phenotypic identification using macroscopic and microscopic appearance. The Non transcribed spacer (NTS) region in
rDNA cluster consist of three subrepeat elements (SREs), TmSO, TmSl and TmS2 which were amplified using Polymerase
Chain Reaction (PCR). Amplification products from each of these three loci showed different types of strain-specific
polymorphism. Combining all these three SREs loci PCR patterns generated 15 individual strain profiles. The PCR fingerprint
of SREs of NTS region in the T. mentagrophytes represents a valuable molecular typing method for future epidemiological
investigations.
