Abstract:

Aspergillus niger is an industrially important organism for the production of enzymes like hemi8208;cellulases and
pectinases. The xylan8208;backbone monomer D8208;xylose is known as an inducing compound for the expression of a
large number of xylan8208;degrading enzymes in A niger.  
In our previous work detailed studies were done on the genes that are part of the XlnR regulon and the way they
are regulated. Now we have moved into a research direction in which we want to use our knowledge to model the
XlnR regulon. With this model we aim to obtain a better understanding of the behavior of the regulon.  Also the
model allows us to quantify and predict responses of the genes involved.  
As a starting point, we hypothesized a model based on our previous regulatory studies. D8208;xylose is used as an
inducer of the XlnR regulon. The model includes the effect of CreA repression by D8208;xylose, having a CreA mutant as
a reference strain. High quality data8208;sets were obtained by qPCR for 22 genes of the regulon from fermentor
grown mycelium having sorbitol as a non8208;inducing/non8208;repressing carbon source and low and high D8208;xylose
concentrations as an inducer.  
In our presentation we link our gene regulation studies with a mathematical model describing the XlnR regulon.
Next, the model structure and the experimental set8208;up are explained. Finally, an interpretation of the data is given
and how it results in reformulating and improving the model.
