Comparative proteomics of tear film from fungal keratitis patients

Abstract:

The study was undertaken to analyse the (i) Tear proteome profile of Aspergillus and Fusarium keratitis patients, (ii)
identification and validation of stage specific biomarkers for mycotic keratitis and (iii) to compare the tear film proteome of
keratitis patients suspected as Aspergillus flavus and Fusarium spp. We have shown earlier the protein profile of tear from
Fusarium sad Aspergillus sp., infected keratitis patients. Among the tear proteins, a fungal glutaredoxin-related protein was
also identified only in Fusarium and Aspergillus infected tear samples. Against this background investigation was made for
some stage specific markers in the A. flavus infected tear. Fluorescence difference gel electrophoresis (2D DIGE) was used to
quantify differential expression of proteins. Mass spectrometry was used for protein identification and differential expression
in Fusarium and A. flavus infected tear samples. 2 glycoprotein (ZAG) was down regulated in A. flavus infected tear. In
Fusarium infected tear samples six proteins were up-regulated when compared with uninfected tear. A similar expression
profile was observed in A. flavus infected tear samples. However, Zinc- -2 glyoprotein (ZAG) is a novel adipokine which is
similar to MHC class I antigen-presenting molecule. The exact physiological function of ZAG in tear is unknown. Macrophage
response is the primary host response to mycotic keratitis and it has been previously shown that macrophage-associated
inflammation plays a significant role in the down regulation of ZAG in adipose tissue. However, the use of ZAG as a biomarker
still needs to be validated with more number of samples.
