Abstract:

A fast and simple technique to analyze a large amount of samples is required for investigation
of microbial ecology. Matrix Assisted Laser Desorption Ionization-Time of Flight Mass Spectrometry
(MALDI-TOF MS) is a powerful analytic tool with advantages including high throughput, high detection
sensitivity and relatively low cost and is frequently used for protein analysis. Several studies have
reported the efficiency of MALDI-TOF MS for the identification of bacterial species but few reports about
fungi are found. In the present study, MALDI-TOF MS was applied to the species characterization of
fungi especially wood degrading basidiomycetes to obtain spectrum pattern for each fungal strain. The all
proteins extracted from fungal culture were analyzed by MALDI-TOF MS. MS spectra were obtained and
compared between intra- and inter species. Results show that some fungal groups are identifiable with
species level and others are identifiable with genus.
