Characterisation of constitutive promoters of Aspergillus niger exhibiting different expression intensities as a useful tool for metabolic engineering

Abstract:

The tool box for the genetic engineering of filamentous fungi has been significantly expanded in the last years.
However, many tools such as promoters of different strength, providing constitutive expression of the target genes
on varying expression levels, are still sparse and not available with a sufficient degree of variability.
Therefore, we set out to isolate and characterize novel constitutive promoters for filamentous fungi providing
both, low and high expression of the target genes. They were characterised with the reporter system, based on the
β 8208;glucuronidase (gusA) activity. Expression cassettes, containing the gusA gene from Escherichia coli controlled by
six novel promoters, were transformed to Aspergillus niger and integrated into the genome. Gene expression from
the different promoters could then be monitored by a sensitive fluorometric assay. Six selected promoters
demonstrated diverse expression levels of the gusA reporter gene in Aspergillus niger.  β 8208;glucuronidase enzyme
activity of the analysed transformants differed in a range of several orders of magnitude. Such a set of constitutive
promoters is a powerful tool for genetic and metabolic engineering purposes, for instance the fine tuning of
metabolic pathways for the improvement of industrial production processes.
To prove functionality of the promoters, we applied them in the metabolic pathway for itaconic acid production.
