I agree with both Paul and Ed. But a couple of things worth adding. You can use sterile gauze from any chemist will probably be cheaper (watman filters are expensive) and quicker (no need to wait on an order) for filtering.

When you aggitate the sporing culture to gain a suspension I find running a sterile cotton swab gently run over the surface works best, but avoid rubbing hard (which would lift more hyphae).

It depends on the application, but as a general rule of thumb I would always store down as Ed mentions at -80oC in glycerol for long term usage, and subculture out fresh each time (using subcultures for up to 2 weeks).

Just treat niger as you would any other Aspergillus species.
